&lt;P&gt;Fecal metabolomics and lipidomics capture integrated molecular signatures of host, diet, and gut microbiome, but traditionally require separate extraction and Liquid Chromatography-Mass Spectrometry (LC-MS) workflows. To develop a unified workflow, multivariate optimization across extraction chemistry, LC-MS conditions, and automation was performed using a Design of Experiments approach, resulting in a scalable automated biphasic extraction and dual UHPLC‑HRMS platform.&lt;/P&gt;
